AdvanceBio Gly-X Technology
AdvanceBio Gly-X technology is a new generation of N-glycan sample preparation. Be ready for sample analysis faster with rapid (5-minute) enzymatic release of N-glycans and labeling with InstantDyes including the brightest in class InstantPC, or traditional reductive amination dyes. Innovative in-solution workflows combined with plate-based rapid cleanup enables sample prep in as little as 1 hour. AdvanceBio Gly-X Increases analytical sensitivity with a variety of dye options, while increasing sample throughput.

Features
- Three dyes are currently available for the Gly-X platform:
- InstantPC: a novel instant N-glycan label for liquid chromatography separations that provides markedly increased MS and HILIC-FLR sensitivity. Sample prep time: ~1 hour.
- 2-AB Express: 2-AB (2-aminobenzamide) has been used to generate labeled N-glycan data for over 20 years and is well established in many laboratories. Traditionally, labeling with 2-AB requires the sample to be dried down prior to the labeling reaction, a step that has been eliminated with Gly-X 2-AB Express on-matrix labeling. Sample prep time: ~2 hours.
- InstantAB: ProZymes original InstantDye which allows room temperature, instant labeling of released N-glycans. Superseded by InstantPC, which has higher fluorescence and MS signal (see Agilent publication 5994-0942EN). Sample prep time: ~1 hour.

Quick and Efficient Sample Cleanup for Biomolecule Analysis
Learn how to use centrifugation with gel filtration cartridges for sample cleanup. This approach is a rapid, convenient, straight-forward technique that can be used to easily avoid the problems associated with analysis of formulated monoclonal antibodies.- 01 Apr 2024
Professor Michael Butler, head of the National Institute for Bioprocessing Research and Training (NIBRT) Cell Technology Group (CTG), explains that the characterisation and analysis of glycans is important for the biopharma sector as it can help in the development of therapeutic agents, such as monoclonal antibodies and other recombinant proteins. These are used in therapies in a range of serious conditions including cancer, inflammatory diseases, and cardiovascular diseases. The mechanism of action depends upon their strong affinity to molecular targets associated with disease.
